    Get PDF

Comparison of 2D and 3D prediction models for environmental vibration induced by underground railway with two types of tracks

Two-dimensional (2D) and three-dimensional (3D) prediction models for environmental vibration induced by underground railway with direct fixation track and steel spring floating slab track are developed and verified. The responses of ground surface calculated by 2D prediction models with various equivalent forces are compared to those calculated by 3D prediction models. The numerical results show that (a) the computational time for each case calculated by 2D prediction models is more than 500 times less than that calculated by 3D prediction models, however, the accuracy of 2D prediction models is relatively lower than 3D prediction models, so 3D prediction models are required for absolute prediction due to their higher accuracy and applicability to a wider range of complex problems; and (b) a suitable equivalent force transfer method for 2D prediction models can improve the prediction accuracy of 2D prediction models, the equivalent forces in 2D prediction models are respectively recommended to use the equivalent wheel-rail force and the equivalent steel spring force averaged over a vehicle length for underground direct fixation track and steel spring floating slab trac

Non-invasive in vivo quantification of the developing optical properties and graded index of the embryonic eye lens using SPIM.

Graded refractive index lenses are inherent to advanced visual systems in animals. By understanding their formation and local optical properties, significant potential for improved ocular healthcare may be realized. We report a novel technique measuring the developing optical power of the eye lens, in a living animal, by exploiting the orthogonal imaging modality of a selective plane illumination microscope (SPIM). We have quantified the maturation of the lenticular refractive index at three different visible wavelengths using a combined imaging and ray tracing approach. We demonstrate that the method can be used with transgenic and vital dye labeling as well as with both fixed and living animals. Using a key eye lens morphogen and its inhibitor, we have measured their effects both on lens size and on refractive index. Our technique provides insights into the mechanisms involved in the development of this natural graded index micro-lens and its associated optical properties

Acute myeloid leukemia in children and adolescents in Brazilian institutions : reality and challenges.

Objective: To describe the outcome of acute myeloid leukemia (AML) among children treated in Brazilian institutions. Methods: A structured online questionnaire was sent to pediatric oncologists affiliated to the Brazilian Society of Pediatric Oncology. The physicians and institutions were unidentified. Results: One hundred and four pediatric oncologists in all Brazilian regions answered the questionnaire. The treatment-related mortality rate was reported to be higher than 30% by 29.8% of the participants. Difficulty in accessing the intensive care unit (ICU) was reported by 54.8%. About 85% had access to cytogenetics, 78% to molecular testing, 94% to the measurement of residual disease by flow cytometry. About 90% of participants reported access to HSCT, but 86% of them had difficulties in providing HSCT timely. About 95% of the respondents indicated the need to create a national treatment protocol, and 89.4% are willing to collaborate with a national study group. Conclusion: Our study demonstrated large gaps in the treatment of pediatric AML in Brazil. To improve outcome, a national protocol will have to consider the regional differences and adapt the management according to the local resources

Automated Reporter Quantification In Vivo: High-Throughput Screening Method for Reporter-Based Assays in Zebrafish

Reporter-based assays underlie many high-throughput screening (HTS) platforms, but most are limited to in vitro applications. Here, we report a simple whole-organism HTS method for quantifying changes in reporter intensity in individual zebrafish over time termed, Automated Reporter Quantification in vivo (ARQiv). ARQiv differs from current “high-content” (e.g., confocal imaging-based) whole-organism screening technologies by providing a purely quantitative data acquisition approach that affords marked improvements in throughput. ARQiv uses a fluorescence microplate reader with specific detection functionalities necessary for robust quantification of reporter signals in vivo. This approach is: 1) Rapid; achieving true HTS capacities (i.e., >50,000 units per day), 2) Reproducible; attaining HTS-compatible assay quality (i.e., Z'-factors of ≥0.5), and 3) Flexible; amenable to nearly any reporter-based assay in zebrafish embryos, larvae, or juveniles. ARQiv is used here to quantify changes in: 1) Cell number; loss and regeneration of two different fluorescently tagged cell types (pancreatic beta cells and rod photoreceptors), 2) Cell signaling; relative activity of a transgenic Notch-signaling reporter, and 3) Cell metabolism; accumulation of reactive oxygen species. In summary, ARQiv is a versatile and readily accessible approach facilitating evaluation of genetic and/or chemical manipulations in living zebrafish that complements current “high-content” whole-organism screening methods by providing a first-tier in vivo HTS drug discovery platform

Attention to pain! A neurocognitive perspective on attentional modulation of pain in neuroimaging studies

    Get PDF
Several studies have used neuroimaging techniques to investigate brain correlates of the attentional modulation of pain. Although these studies have advanced the knowledge in the field, important confounding factors such as imprecise theoretical definitions of attention, incomplete operationalization of the construct under exam, and limitations of techniques relying on measuring regional changes in cerebral blood flow have hampered the potential relevance of the conclusions. Here, we first provide an overview of the major theories of attention and of attention in the study of pain to bridge theory and experimental results. We conclude that load and motivational/affective theories are particularly relevant to study the attentional modulation of pain and should be carefully integrated in functional neuroimaging studies. Then, we summarize previous findings and discuss the possible neural correlates of the attentional modulation of pain. We discuss whether classical functional neuroimaging techniques are suitable to measure the effect of a fluctuating process like attention, and in which circumstances functional neuroimaging can be reliably used to measure the attentional modulation of pain. Finally, we argue that the analysis of brain networks and spontaneous oscillations may be a crucial future development in the study of attentional modulation of pain, and why the interplay between attention and pain, as examined so far, may rely on neural mechanisms shared with other sensory modalities
